No, Takoizu "died", but because of the fact that she is of Youkai descent, she simply regenerated. She can only truly die by magical and legendary weapons, just like any other Youkai. The only difference is that she is not as durable as a Youkai and does not have indestructible bones like John.

Exactly. She's essentially a "glass cannon", meaning that she is incredibly dangerous, but she is susceptible to damage.
Her bones are strong enough to block .45 caliber rounds, but any higher and the bullets pass right through. Her bones are tough, but not quite as tough as John's. Unlike Yukari, who is highly resistant to non-magical damage, Takoizu is affected just like John. Not only that, she also has all the weaknesses of a Youkai, such as being weak to seals, magical weaponry, and spiritual-based attacks.
